Résumé

En tant qu’acteur clef de l’axe cérébro-hypohysogonadique, la GnRH (Gonatropin-Releasing Hormone) a fait l’objet, essentiellement chez les Vertébrés, d’un nombre considérable de recherches. Des études biochimiques, moléculaires, neuroanatomiques, pharmacologiques et physiologiques, principalement chez les Mammifères, mais aussi chez les Poissons, ont conduit à une connaissance détaillée des systèmes à GnRH hypophysiotropes et de la GnRH en tant que facteur de libération des gonadotrophines hypophysaires. Il est à présent admis que les neurones à GnRH se développent à partir de l’épithélium olfactif et migrent, au cours de l’organogenèse, dans le cerveau antérieur pour établir des connexions avec l’éminence médiane chez les Tétrapodes ou l’hypophyse chez les Poissons Téléostéens. Cependant, tous les Vertébrés possèdent un second système à GnRH, exprimant dans des neurones du synencéphale un variant connu sous le nom de chicken GnRH-II dont les fonctions sont méconnues. De plus, de nombreuses espèces de Poissons Téléostéens présentent une troisième forme, le salmon GnRH, dont l’expression est limitée à des neurones des systèmes olfactifs et du télencéphale ventral projetant largement dans l’ensemble du cerveau et à un degré moindre vers l’hypophyse. Chez les Vertébrés, des peptides de type GnRH sont également exprimés dans les gonades où ils agissent sur la prolifération cellulaire ainsi que la stéroïdogenèse chez les mâles et sur la réinitiation de la méiose et l’apoptose de la granulosa chez les femelles. Il est possible que de telles fonctions représentent l’un des rôles ancestraux de la GnRH puisqu’un nombre croissant d’études chez les Invertébrés met en évidence des connexions plus ou moins directes entre des neurones sensoriels et les gonades. D’après ces études, la GnRH serait un peptide très ancien qui aurait émergé au plus tard chez les Cnidaires, considérés comme les premiers animaux possèdant un système nerveux. Outre les Cnidaires, des peptides de type GnRH ont été caractérisés partiellement chez quelques classes d’invertébrés tels que les Mollusques, les Echinodermes et les Prochordés chez lesquels des effets de la GnRH sur la reproduction, notamment la libération des gamètes ou la stéroïdogenèse, ont été décrits. Il est possible qu’avec la complexité croissante des Métazoaires, les neurones à GnRH aient perdu leurs connexions directes avec la gonade et se soient spécialisés dans le contrôle de nouveaux centres de régulation, tels que l’hypophyse chez les Vertébrés ou la glande optique chez les Céphalopodes. Néanmoins, les fonctions de la GnRH au niveau gonadique auraient été conservées grâce à une production locale de peptides. L’ensemble de ces données suggère que la GnRH était impliquée dans le contrôle de la reproduction avant l’apprition des cellules gonadotropes hypohysaires.

Abstract

As a major actor of the brain-pituitary-gonad axis, GnRH has received considerable attention, mainly in vertebrates. Biochemical, molecular, neuroanatomical, pharmacological and physiological studies have mainly focused on the role of GnRH as a gonadotrophin-releasing factor and have led to a detailed knowledge of the hypophysiotrophic GnRH system, primarily in mammals, but also in fish. It is now admitted that the corresponding neurons develop from the olfactory epithelium and migrate into the forebrain during embryogenesis to establish connections with the median eminence in tetrapods or the pituitary in teleost fish. However, all vertebrates possess a second GnRH system, expressing a variant known as chicken GnRH-II in neurons of the synencephalon, whose functions are still under debate. In addition, many fish species express a third form, salmon GnRH, whose expression is restricted to neurons of the olfactory systems and the ventral telencephalon, with extensive projections in the brain and a minor contribution to the pituitary. In vertebrates, GnRHs are also expressed in the gonads where they act on cell proliferation and steroidogenesis in males, and apoptosis of granulosa cells and reinititaion of meiosis in females. These functions could possibly represent the primitive roles of GnRH-like peptides, as an increasing number of studies in invertebrate classes point to a more or less direct connection between GnRH-producing sensory neurons and the gonads. According to recent studies, GnRHs appear as very ancient peptides that emerged at least in the cnidarians, the first animals with a nervous system. GnRH-like peptides have been partially characterized in several classes of invertebrates notably in molluscs, echinoderms and prochordates in which effects on the reproductive functions, notably gamete release and steroidogeneis, have been evidenced. It is possible that, with the increasing complexity of metozoa, GnRH neurons have lost their direct connection with the gonad to specialize in the control of additional regulatory centers such as the hypophysis in vertebrates or the optic gland in cephalopods. However, reminiscent effects of GnRH functions at the gonadal level would have persisted due to local production of GnRHs in the gonad itself. Altogether, these data indicate that GnRHs were involved in the control of reproduction long before the appearance of pituitary gonadotrophs.
